Pathological protein fibril formation can be observed in chronic neurodegenerative diseases such as Alzheimer’s disease (AD), Parkinson’s disease (PD), Huntington’s disease (HD) and prion disease.(1) AD is characterized by the aggregation of amyloid β peptides (Aβ) into fibrillar amyloid plaques in selected areas of brain.(2) A number of efforts in searching for inhibitors of Aβ aggregation has demonstrated that small molecules, containing single or multiple (hetero)aromatic rings, often bearing hydroxy groups, can inhibit protein-protein interactions.(3) A not exhaustive list of these structurally diverse inhibitors includes indole derivatives, such as polysubstituted fluorinated indoles, melatonin, hydroxyindoles, as well as polyphenols, such as curcumin and resveratrol.
