A method for its detn. in urine samples by HPLC with indirect anodic polarog. detection was developed. The method is simple and highly selective and permits the direct injection of urine after diln. (1:20) with mobile phase. Creatinine, which also gave an anodic response on mercury, could be simultaneously detd. and used as an internal std. for pseudouridine. This allowed detn. of the pseudouridine/creatinine ratio in a single run on the same specimen and the use of urine samples collected randomly instead of a 24-h collection. The within-day coeff. of variation for pseudouridine/creatinine peak area ratios was 3.4%.
